In Wisconsin, US Route 10 spans the width of the central part of the state of Wisconsin, running eastward from Prescott to Manitowoc. Prescott is just across the Mississippi River from Hastings, Minnesota, and US Route 10 crosses the bridge connecting these two communities as it enters Wisconsin. From there, US Route 10 moves eastward, staying south of Interstate 94 and Eau Claire. Interstate 94 begins to curve southeasterly around here, and US Route 10 crosses this southeastward path at Osseo. US Route 10 continues east from here, serving Marshfield and Stevens Point, where US Route 10 meets Interstate 39 and US Route 51.  US Route 10 now ventures into the eastern half of Wisconsin, acquiring a southeasterly angle. US Route 10 is mostly a freeway from Stevens Point to Appleton, where US Route 10 meets Interstate 41 (US Route 41). By Appleton, US Route 10 cuts south from its freeway (which becomes Wisconsin 441), and it turns eastward through Brillion. As US Route 10 approaches Lake Michigan, it is swept up by Interstate 43 and carried southward to a different east-west road bound for the north side of Manitowoc. Just before the lakeshore, US Route 10 jumps south onto Wisconsin Highway 42 into the heart of Manitowoc. US Route 10 meets a ferry dock, where US Route 10 is suspended, and the route reappears on the east side of Lake Michigan in Ludington, Michigan, at the opposite ferry dock.
