In Wisconsin, US Route 151 cuts a northeast-southwest path from Manitowoc to the very southwest corner of the state. The northeastern end of US Route 151 is at Interstate 43 on the southwest side of Manitowoc. The east-west road, where US Route 151 manifests, carries Wisconsin Highway 42, which splits off toward the south after a quarter of a mile. US Route 151 continues westward along its first stretch, bound for Chilton and the eastern shore of Lake Winnebago. The lake comes within view, and the road bends to the left (south) to hug the eastern shore. Another road appears and goes north along the eastern shore; that road is Wisconsin Highway 55. Regardless, US Route 151 wraps around the southeast side of the lake, reaching Fond du Lac. US Route 151 meets US Route 45 and Interstate 41, and it continues southwestward as a four-lane expressway. That expressway is bound for Madison, but it serves Waupun, Beaver Dam, Columbus, and Sun Prairie before getting there. US Route 151 gives up the whole expressway thing when it crosses the triplicate Interstate Highway (39/90/94), because it becomes the sole numbered highway traversing the isthmus through downtown Madison. US Route 151 follows Washington Avenue for the most part. However, it no longer goes straight past the capital, because it was rerouted south to John Nolen Drive in the heart of downtown. It does jump back to Washington Avenue before continuing south out of Madison. When it reaches the Beltline carrying US Routes 12 and 18 , it joins the Beltline west for a short while. Then, US Route 151 follows US Route 18 as that route splits from the Beltline to continue southwestward. US Routes 18  and 151 run together on a freeway all the way until Dodgeville. US Route 18 leaves the freeway, and US Route 151 has the freeway all to itself. The freeway becomes more of an expressway, though, passing by Belmont and Dodgeville. This expressway for US Route 151 lasts all the way until it leaves the state into Dubuque, Iowa. US Route 61 uses the same bridge over the Mississippi River to enter Iowa.
