Indiana State Route 159 is a designation for three separate segments of north-south roadway. The northernmost is from Indiana Route 46 at Riley to Indiana Route 48 at Jasonville. The second segment is a spur from Indiana Route 54 in Dugger south to Pleasantville. The southernmost segment is from Indiana Route 58 in Freelandville south to Indiana Route 67 in Bicknell, a town northeast of Vincennes.
