Indiana State Route 340 is an east-west route that runs for five and a half miles just west of Brazil, Indiana. This is just east of the Terre Haute area. Indiana Route 340 is an alternate route to US Route 40, running on a former alignment of the US highway that is never more than one-third of a mile from the current one. Indiana Route 340 starts at US Route 40 at Cloverland, runs east paralleling US Route 40 to the north by a third of a mile at most, then meets US Route 40 again on the west side of Brazil.

The western end of Indiana Route 340 is also the south end of another, unmarked, road, and this sign assembly faces south toward US Route 40 to specify that the state highway is to the right of this fork.
