In Indiana, US Route 50 runs east-west to span the width of the southern part of the state. Specifically, US Route 50 runs from Vincennes to the Cincinnati area while crossing Indiana. As soon as it finishes crossing the Wabash River out of Illinois, US Route 50 meets US Route 41, which drops US Route 150 onto US Route 50. US Routes 50 and 150 continue eastward together through a quarter of Indiana's width, until the town of Shoals. US Route 50 serves as the main east-west thoroughfare in Seymour, Indiana, where it meets Interstate 65 and US Route 31. US Route 50 passes through Aurora and Lawrenceburg in Indiana's southeast corner, before crossing into Ohio and the Cincinnati area.

The sign assembly depicted faces north, at traffic oriented southward at Indiana Route 446's southern end.
