In Ohio, US Route 35 runs from west-central Ohio to southeastern Ohio, passing through Dayton and Chillicothe along the way. Like US Route 33, it is a diagonal route that, overall, runs northwest to southeast. US Route 35 enters Ohio by using Interstate 70 east out of Richmond, Indiana. As soon as Interstate 70 reaches Ohio, it tells US Route 35 to quit mooching and find its own way toward the east. Interstate 70's first exit in Ohio sends US Route 35 toward the southeast so it can reach Eaton. Next, US Route 35 continues east through several more towns. Soon, it becomes a freeway through downtown Dayton and beyond. US Route 35 bypasses Xenia to the south, and it turns southeasterly, remaining as a freeway. US Route 35 continues beyond Interstate 71 and Washington Court House to join US Route 23's freeway in Chillicothe. The two routes join together moving southerly for a short period. Soon, US Route 35 splits away to continue southeast. The freeway character soon subsides, but US Route 35 remains a rural high-speed 4-lane highway for the rest of its length in Ohio, passing Jackson and entering Gallia County. US Route 35's last course of action in Ohio is to pass around the north side of Gallipolis, connecting to Ohio Route 7 via Ohio Route 735. US Route 35 exits Ohio on a bridge over the Ohio River into Henderson and Point Pleasant, West Virginia.

Drivers encountering the junction marker in the photo are moving northeasterly toward a diamond interchange with US Route 35, which forms an expressway bypass around the northeast side of Jackson.
