Ohio State Route 159 connects Lancaster to Chillicothe in the southern areas of central Ohio. As Lancaster is northeast of Chillicothe, the route runs northeast to southwest. The northeastern end is at US Route 22, just barely west of US Route 33 and the Lancaster city limits. Ohio Route 159 goes southwesterly from there to Kingston, where the route becomes more southerly for its final stretch. The route nudges to the western side of US Route 23 before ending in the heart of Chillicothe, at US Route 50 and Ohio Route 104.
